PhD Thesis in Biology, microbiology: "Characterisation of higBA, a Toxin-Antitoxin system of Caulobacter crescentus".
Creation, management and implementation of a scientific research project.
Understanding of the function of a genetic system, called Toxin-Antitoxin, in the resistance of bacteria to antibiotics.
Use of various scientific techniques:
Biochemistry: total extraction of bacterial proteins, Western blot. Total extraction of bacterial RNA and DNA. Measure of the enzymatic activity of Beta-galactosidase.
Molecular biology: gene insertion in a plasmid reporter, transformation and transduction of bacteria. Chromatine Immuno-Precipitation followed by quantitative PCR (Q-ChIP), or by deep sequencing of the genome (ChIP-SEQ). Transposon insertion followed by deep sequencing of the genome (Tn-SEQ).
Bioinformatical analyses: analysis of ChIP-SEQ and Tn-SEQ, use of scientific softwares to transform and analyse deep sequencing datas.
Cellular biology: Bacterial cultures. Synchronization of bacterial cultures. Stress induction on bacterial cultures. Infection using phages. Construction of mutant bacterial strains by gene deletion.
First thesis project in the study of insulin production. Characterisation of a cell line able to produce high quantity of insulin.
